Dr. Dhruv Sareen's independent research laboratory at the Cedars-Sinai Medical Center, Regenerative Medicine Institute focuses on using human iPSC technology to understand the molecular mechanisms underlying neurodevelopmental degenerative diseases and how obesity is linked to the brain. His laboratory is particularly interested 'disease-in-a-dish' modeling for neuromuscular. neurodegenerative and motor neuron diseases including, amyotrophic lateral sclerosis (ALS). They are particularly interested in C9orF72 and other genetic forms of ALS and what are the common elements with sporadic forms of ALS. Using the iPSC platform his lab has developed novel methods for drug screening on patient motoneurons and dissecting mechanism for motoneuron degeneration. In the context of stem cell modeling of neurodevelopmental (SMA) and neurodegenerative diseases (ALS) Dr. Semen's group is particularly interested in the mechanistic insights how specific neural cell types degenerate during the disease processes. They are using ’OMICS (proteome, transciptome, metabolome) and imaging methodologies to gain mechanistic insights into the mitochondrial and endoplasmic reticulum stress involvement. In addition, by understanding how the cells of the nervous system develop and differentiate we will be able to develop better and safer cells for transplantation and regenerative therapies. Dr. Smelt's group has extensive experience in pluripotent and neural stem cell research, He has developed and directed a well-renowned human iPS cell core, He has published high-profile manuscripts on luau iPSC-based disease modeling in SMA and ALS.

Dr. Sareen's laboratory is searching for a highly-capable Post-doctoral Researcher to join his research group to investigate disease mechanisms in neurodegenerative diseases, especially motor neuron disorders. The incumbent should have experience in in neurobiology, biochemistry, molecular biology and stem cells, including analyzing their genetic and biochemical profiles by OMICS technologies. Bioinformatics (programming in R) knowledge is a plus. By using human iPS cell technology and differentiating into different brain cells as well as other tissues of the body we will investigate brain-linked disease mechanisms to develop assays in patient-derived neural cell types and screen for novel therapeutics. Working independently but in close cooperation and in consultation with the Pl and other Research Scientists at RMI, the Post-doctoral Researcher will perform routine and complex laboratory procedures throughout. Tice Post-doctoral Researcher will be expected to assist the PI in preparation of multiple grant proposals and submission of such proposals. They will also prepare full manuscripts for publication as primary first and co-authors. The researcher will be expected to assist the PI in maintaining institutional IRB/SCRO protocols as well as develop new animal and IACUC protocols. The incumbent will develop, adapt, and implement new research techniques and protocols and will analyze and interpret data. They will participate in presentations as author or co-author on scientific meeting abstracts.

Under the direction of Dr. Sareen (FI) the incumbent will help us achieve our re-search goals by performing the principal duty of using pluripotent and neural stem cell culture techniques to generate neuronal and other tissue differentiation protocols, characterization of the differentiated cells (control vs disease) using techniques such as gene and protein expression, physiological. and phenotypic assessments. The candidate will also be expected to develop and perform differentiation protocols to direct pluripotent stem cells to endoderm, ectoderm and mesoderm lineages. The successful candidate will be responsible for all cell maintenance, expansion and genetic modification using viral vectors or other gene-editing methods. Therefore, experience in stern cell and neurobiology, knowledge of pluripotent neural cell markers combined with a proven record in stem cell culture techniques is essential. Experience with bioinformatics analysis of RNA-seq and mass spec proteotnics data are important.

Prior experience in novel genome-editing technologies (CRISPRICas9) using pluripotent stem cells is a great asset, Prior experience in high-content compound screening/imaging techniques. handling of the Molecular Devices Image Express Micro system, and a Mass Spectrometer with data analysis are preferred traits. Proficiency in computer algorithm development for automating image-based analysis is also a desirable trait.

ESSENTIAL JOB DUTIES AND RESPONSIBILITIES:

Assists in preparation of grant proposals and institutional protocols.

Participates in publications and presentations as author or co-author.

Designs and performs experiments. Will keep appropriate experimental records and documentation and analyze the results with the Principal Investigator.

May develop, adapt, and implement new research techniques and protocols.

Analyzes, interprets, summarizes and compiles data.

Performs routine and complex laboratory procedures throughout training period.

Operates and maintains equipment and instruments.

Participation in laboratory meetings, keeping a lab notebook, performing calculations, tabulating data, summarizing methods and results of related experiments and general lab maintenance will he required. The individual will be expected to function efficiently and cooperatively with a team of faculty, research associates, technicians and students.

Additional Information:

Scheduling flexibility including evening, weekend and holiday rotation commitment is requited, Excellent written and oral communication skills are essential. Travel may be required.

Educational Requirements:

Doctorate (MD, PhD, VMD, or DDS) in area directly related to field of research specialization.

Experience:

Interested candidate should be within 0-2 years after PhD attainment. Independently works on research projects designed by a mentor (typically the PI) within an area of specialization. Acquires thorough technical and theoretical knowledge of research project and objectives during one to five (1-5) year post-doctoral appointment, Demonstrated aptitude to perform experimental protocols and procedures, including detailed data collection, and analysis and operation and maintenance of specialized equipment, Knowledge of safety standards and maintenance of specialized equipment.
